What are peptides? A research-use-only overview

A peptide is a short chain of amino acids joined together in a defined order. Amino acids are small organic molecules, each with a common backbone and a distinguishing side chain, and they are the same building blocks that assemble into proteins. The practical difference between a peptide and a protein is length: peptides are generally understood to be the shorter chains, while proteins are the longer, often folded assemblies. There is no universal cutoff, but many references describe chains of up to a few dozen amino acids as peptides.

Amino acids and the peptide bond

Each amino acid carries an amino group at one end and a carboxyl group at the other. When two amino acids join, the carboxyl group of one reacts with the amino group of the next, releasing a water molecule and forming a covalent linkage called a peptide bond. Repeating this reaction produces a chain with a free amino end (the N-terminus) and a free carboxyl end (the C-terminus). By convention, a peptide sequence is written and numbered from the N-terminus to the C-terminus, which is why documentation and reference literature list residues in that direction.

Sequence and identity

The order of amino acids in the chain is the peptide sequence, and it is the primary way a peptide is identified. Because there are twenty common amino acids, even a short chain represents an enormous number of possible sequences, and a change to a single position produces a distinct molecule. Reference materials frequently note the sequence alongside a molecular formula and a molecular weight, giving three independent descriptors of the same molecule. Analytical documentation such as a Certificate of Analysis exists to confirm that a supplied material matches its stated identity.

A peptide is commonly described by three linked descriptors: its amino acid sequence, its molecular formula (the count of each element), and its molecular weight. Analytical methods are used to check that a material is consistent with all three.

How peptides are studied

In a research setting, peptides are handled as defined chemical materials. Investigators characterize them using analytical instruments: chromatography separates a sample into its components so that identity and purity can be assessed, and mass spectrometry measures molecular mass to confirm that the observed value matches the expected sequence. Synthetic peptides are commonly supplied as a lyophilized (freeze-dried) powder, a stable form suited to storage and laboratory reference. These are the kinds of technical descriptors — purity, mass, appearance, and lot identity — that appear in a research catalog record rather than any account of what a material does.
